Chenodeoxycholic acid and ursodeoxycholic acid reduce bioavailability of nitrendipine

In a single-dose study,6 healthy subjects were given nitrendipine 10mg with or without either chenodeoxycholic acid 200mg or 600 mg, or ursodeoxycholic acid 50 mg. Ursodeoxycholic acid reduced peak plasma level and AUC of nitrendipine by 54 % and 75%, respectively. Chenodeoxycholic acid 200mg decreased peak plasma level and AUC of nitrendipine by about 20%, but 600mg dose reduced peak plasma level and AUC of nitrendipine by 54 % and 68%, respectively. The reduction in bioavailability of nitrendipine was possibly due to effects of bile acids on tablet disintegration or more probably on drug solubilisation.
